Aikurō Mikisugi is introduced as Ryuko Matoi's homeroom teacher at Honnouji Academy. He is secretly an undercover agent of the anti-Life Fiber paramilitary organization Nudist Beach. After Ryuko stumbles across the sentient sailor uniform Senketsu beneath the ruins of her father's home, Mikisugi reveals his true allegiance and begins aiding her. He provides information about Life Fibers and the true nature of the threat they pose. Later, Ryuko learns that her father Isshin Matoi founded Nudist Beach and created Senketsu to fight the Life Fibers, which are actually parasitic aliens that prompted humanity's evolution to later feed on them. Mikisugi's role as a teacher and agent places him at the center of the conflict between Ryuko, Satsuki Kiryuin, and Ragyo Kiryuin's fashion empire.

In Their Own Story

Mikisugi first appears as Ryuko's homeroom teacher, but after her failed attempt to challenge Satsuki on her first day, he reveals himself as an undercover agent of Nudist Beach. He guides Ryuko through her discovery of Senketsu and the truth about Life Fibers. When Ryuko and Nudist Beach storm the Honnouji Academy festival held in Ragyo's honor, Mikisugi fights alongside them. After Ragyo and the Life Fibers devastate Japan, the Elite Four and Nudist Beach rescue both Satsuki and Mako. Mikisugi is part of this rescue effort. In the final battle, Ryuko pursues her mother into space after everyone offers their Goku Uniforms to power up Senketsu. Following the victory, in the OVA episode set two weeks later, Rei Hououmaru creates doppelgängers of Satsuki and the Elite Four, which are defeated. Honnō City and the school sink into Tokyo Bay, and everyone can finally live a normal life free of the Life Fibers.
